 Hepatocellular carcinoma (HCC) is the most common primary cancer of the liver, accounting for 70% - 85% of liver cancer cases. HCC has the leading incidence and mortality rate of cancer diseases in general. All most HCC cases were detected in late stage and patients have little chance of effective treatments. Recently, the use of microRNA molecules as potential markers has been focused on the diagnosis of HCC cases and other cancer diseases. In this study, the expression levels of miRNAs between HCC and normal liver tissues provided by the microarray dataset collected from Gene Expression Omnibus (GEO) were evaluated by bioinformatics tool. Then, ten candidate microRNA molecules were selected base on previous studies and expression profiles of these miRNAs in plasma samples were evaluated with three groups of HCC, Hepatis B patients and healthy adults using real-time PCR. The results indicated that at least two microRNAs (hsa-miR-192 and hsa-miR-10a) showed a significant difference in the expression between HCC and healthy groups in tissue and plasma samples. In contrast, two microRNA including hsa-miR-193a and hsa-miR-25 showed increased expression in only HBV-infected group.

 

The results of combined ROC curve analysis between miRNAs showed that cluster of hsa-miR-25 and hsa-miR-10a was the best with the average AUC value of the microarray and plasma samples were 96% and 85.7% respectively, which has implications for the non-invasive marker approach and understanding of the expression of potential microRNAs, which promising for developing appropriate tools to diagnosis and prognosis of hepatocellular carcinoma in Vietnamese patients.
