Panax  vietnamensis  Ha  et  Grushv.) is a specific and precious Panax in Vietnam with high economic pharmaceutical value.Embryo numbers by callus treatment, which derived from tubers exhibited higher volume than from leaves. The SH medium was observed to be more suitable to the formation of calli in which embrygenesis taking place potentially. Otherwise, the MS medium became better to be sensitive in case of embryogenesis from Ngoc Linh panax leaf calli.

ABSTRACT

Panax  vietnamensis  Ha  et  Grushv.) is a specific and precious Panax in Vietnam with high economic pharmaceutical value.

Embryo numbers by callus treatment, which derived from tubers exhibited higher volume than from leaves. 

The SH medium was observed to be more suitable to the formation of calli in which embrygenesis taking place potentially. Otherwise, the MS medium became better to be sensitive in case of embryogenesis from Ngoc Linh panax leaf calli.

In the basic MS medium, if 2,4-D was added separately with  NAA or kinetin, the output would not be efficient. However, the MS medium, which associated with 2,4-D, NAA and kinetine, was noticed to be highly effective in embryogenesis. Especially, the dose of 2,4-D, NAA and kinetine was 1.0 mg/L, 0.5 mg/L and 0.2 mg/L, respectvely, would create the highest rate of embryos. Sucrose, glucose and fructose could highly induce embryogenesis at the level of 50 g/L. Especially, sucrose could enhance embryogenesis frequency of 87.6% and embryo formation of 55.7%. Arginine and ornithine did not significantly enhance the frequency in termof Ngoc Linh panax. Spermidine at the dose of 150 µM obtained the optimum output. Under the condition of light treatment of 16 hours per day, the culture of shaked liquid medium became suitable to embryogenesis and callus growth og Ngoc Linh panax. The seedlings from somatic embryogenesis expressed their high survival percentage under nurseries located in Ngoc Linh mountain. They have had the genetic uniformity via RAPD analysis.
