Novel Platform Visually Screens Gene Editing Efficiency in Porcine

Researchers developed a platform to visually screen the efficiency of gene editing in porcine. The platform is a simple, rapid, and effective way to assess gene knockout efficiency.

 

Gene-edited pigs are typically produced using CRISPR-Cas9 and somatic cell nuclear transplantation (SCNT). The efficiency in getting gene-edited nuclear donors is affected by the chosen CRISPR-Cas9 form. However, there is no direct technique to assess the efficiency of CRISPR-Cas9 in porcine.

 

To address this issue, researchers produced a platform based on micropattern arrays and fluorescence signals to visually evaluate gene editing's efficiency. The team compared the editing efficiency of DNA, mRNA, and Ribonucleoprotein (RNP) forms of CRISPR-Cas9. Their findings showed that the mRNA form that targets porcine cells had the highest editing efficiency. The platform also demonstrates great potential for quickly analyzing gene editing tools and delivery methods.
