Abstract

 

Improving photosynthetic efficiency through increasing the efficacy of photosynthetic carbon assimilation and optimizing the source-sink strength is an important aspect to raise the crop yield potential. The overexpression of mesophyll envelope protein 1 (MEP1) which is one of the key transportersinvolved in the C4 photosynthetic pathway will help in engineeringthe C4 photosynthetic pathway into C3 rice to increase the enzyme activities and overcome the minor fluxes.MEP1 from maize was successfully overexpressed in transgenic rice plants. The phenotyping results showed that the leaves of ZmMEP1 plants exhibited pale green color with chlorosis which did not change over generations.The rate of photosynthetic CO2 assimilation was also reduced in these plants as compared to the wildtype. Interestingly, there was also significant low starch and high sucrose content in the leaves indicating that the ZmMEP1 plants rapidly converted starch into sucrose in the metabolites content of the leaves. This might be due to the imbalance in the redox in the pathway as a result of the lack of the other components of the C4 pathway for this transporter to perform its function in the C3 environment. These plant materials are very useful resources to build up an entire C4 biochemical pathway in rice by intercrossing these with other plants over expressing C4 enzymes and transport proteins.
