The MS medium, which was added by 0.5 mg/L of 2,4 D was obtained the best effectiveness to induce calli. The combination between 2,4 D and KIN was noticed to help somatic embryos create calli of jatropha. The basic MS medium, which was complemented by spermidine at the level of 0.03 mg/L, was notice to optimize the embryogenesis. Otherwise, the basis B5 medium, which was complemented by 1.5 mg/L of IBA, was suitable the best to form roots, and seedling growth in vitro. The different phenotypes could be divided into four morphological types as regular type without variation (72.3%), leaf variants, shoot variants and leaf + shoot variants. The variants accounted for 27.7%. Genetic divergence analysis by 19 RAPD markers obtained high similarity at the level of 0.98 among the seedlings after regeneration. It demonstrated their genetic uniformity. The created protocol that could optimize the micropropagation of Jatropha curcas L. was proposed to be applied. The seedlings would be obtained after 49 weeks via the micropropagation with 5 million seedlings / somatic embryo. The seedlings are uniform, free diseases, well rooting to adapt to large scale production for biofuel industry. |
