Increasing grain yield and improving grain quality are two important goals for rice breeding. A better understanding of the factors that contribute to the overall grain quantity and nutritional quality of rice will lay the foundation for developing new breeding strategies. RAG2 is a member of 14-to-16-kDa α-amylase/trypsin inhibitors in rice, which belong to the albumin of seed storage proteins. We found that RAG2 was specifically expressed in ripening seed and its transcription peak was between 14 and 21 days after flowering. Grain size and 1000-grain weight were obviously increased in RAG2-overexpressed lines compared with wild type, and grain size was reduced in RAG2-suppressed lines.

Sequence analysis showed that OsACOS12 shares 63.9 % amino acid sequence identity with ACOS5. The osacos12 mutation caused by a pre-mature stop codon in LOC_Os04g24530 exhibits defective sexine resulting in a male sterile phenotype in rice. In situ hybridization shows that OsACOS12 is expressed in tapetal cells and microspores at the transcript level. The localization of OsACOS12-GFP demonstrated that OsACOS12 protein is accumulated in tapetal cells and anther locules.

We identified the biosynthetic pathway for the nonsugar sweetener mogroside V, a noncaloric with a sweetening strength 250-fold that of sucrose. This compound is produced by the fruit of the endemic Chinese cucurbit Siraitia grosvenoriii, also known as monk fruit and luo-han-guo. The metabolic pathway was identified using a combination of genomic and transcriptomic databases of the Siraitia plant, together with a large-scale functional expression of candidate genes.

Maize (Zea mays L.) is an important food and energy crop, and low phosphate (Pi) availability is one of the major constraints in maize production worldwide. Plants adapt suitably to acclimate to low Pi stress. However, the underlying molecular mechanism of Pi deficiency response is still unclear. In this study, yeast HOG1 homologue from the root endophyte Piriformospora indica (PiHOG1) was isolated and functionally characterized. Functional expression of PiHOG1 in S. cerevisiae ∆hog1 mutant restored osmotolerance under high osmotic stress. Knockdown (KD) transformants of PiHOG1 generated by RNA interference in P. indica showed that genes for the HOG pathway, osmoresponse and salinity tolerance were less stimulated in KD-PiHOG1 compared to the wild-type under salinity stress.

STAT6 is a transcription factor and plays a predominant role in IL-4/IL-13 and virus-mediated signaling pathways. Extensive studies have linked malfunctions of STAT6 to pathological features of asthma and cancer. Targeting the function of STAT6 has become an attractive therapy. Understanding the molecular mechanisms of STAT6 transcriptional regulation is still scarce. Here, we report the atomic-level structures of the phosphorylated STAT6 core fragment homodimer, both in DNA-free and complexed with N4 or N3 site DNA, uncovering both a larger dimer interface intersection angle and the unique residue H415 of STAT6 as important factors for discrimination of N4 from N3 site DNA.
